摘    要:目的:探讨pan-RAF抑制剂LY3009120对甲状腺癌细胞增殖、侵袭和迁移的影响及可能机制。方法:以不同浓度的LY3009120作用于MDA-T32细胞,应用CCK-8法检测细胞的增殖能力。以半数致死浓度(half maximal inhibitory concentration,IC50)作用于细胞,划痕实验检测细胞迁移能力,Transwell实验检测细胞侵袭能力。Western blotting法检测细胞中E-cadherin、N-cadherin和Snail的表达。结果:通过CCK-8法检测显示LY3009120能明显抑制MDA-T32细胞的增殖。划痕实验和Transwell实验结果显示LY3009120能抑制细胞的迁移和侵袭。Western blotting结果显示LY3009120促进MDA-T32细胞中E-cadherin的表达,抑制N-cadherin和Snail的表达。结论:LY3009120能够抑制甲状腺癌细胞系MDA-T32的增殖,可能通过上皮间质转换机制抑制细胞的侵袭和迁移。

Abstract:Objective:To investigate the effects of LY3009120 on the proliferation,invasion and metastasis of thyroid cancer cell lines and its possible mechanisms.Methods:MDA-T32 cells were treated with different concentrations of LY3009120 and cell proliferation was detected by CCK-8.The half maximal inhibitory concentration(IC50) was used to perform wound healing assay and Transwell assay to detect cell migration and invasion abilities.The expressions of E-cadherin,N-cadherin and Snail were detected by Western blotting.Results:LY3009120 suppressed the proliferation of MDA-T32 cells significantly.LY3009120 inhibited MDA-T32 cell invasion and migration.Moreover,LY3009120 increased the expression of E-cadherin but suppressed N-cadherin and Snail expressions.Conclusion:LY3009120 inhibited MDA-T32 cell proliferation,invasion and metastasis maybe through epithelial-mesenchymal transition mechanism.
