后部巩膜F4/80和CD206阳性巨噬细胞在小鼠形觉剥夺性近视发生发展中的变化

摘    要:目的 研究后部巩膜F4/80+和CD206+巨噬细胞在小鼠形觉剥夺性近视发生发展中的变化趋势。方法 将24只(48眼)4周龄C57BL/6小鼠按随机数字表法分别接受形觉剥夺处理和无处理(各12只),研究分为3组:形觉剥夺组(单眼遮盖,n=12),自身对照组(对侧眼无遮盖,n=12),正常对照组(无处理组右眼,n=12)。实验前(0 d)进行屈光度检测;于实验14 d和28 d测量屈光度后分别处死6只小鼠,分离眼球测量眼轴,并分离后部巩膜组织,采用流式细胞术检测后部巩膜组织中F4/80和CD206的表达,观察各指标的动态变化和内在关联。结果 实验0 d,形觉剥夺组、自身对照组和正常对照组的屈光度差异均无统计学意义(均为P>0.05)。实验14 d,3组屈光度和眼轴差异均无统计学意义(均为P>0.05);形觉剥夺组后部巩膜F4/80+巨噬细胞以及F4/80+、F4/80+/CD206+巨噬细胞的百分比均显著高于自身对照组(均为P<0.05)。实验28 d,形觉剥夺组与自身对照组比较,屈光度明显向近视偏移(P=0.001);并伴有眼轴的显著延长(P=0.02);形觉剥夺组后部巩膜F4/80+巨噬细胞以及F4/80+/CD206+巨噬细胞的百分比均显著高于自身对照组(均为P<0.05)。实验28 d,形觉剥夺组小鼠后部巩膜F4/80+/CD206+巨噬细胞的百分比显著高于实验14 d时(P=0.02)。实验28 d,形觉剥夺组后部巩膜F4/80+/CD206+巨噬细胞的百分比和屈光度呈显著负相关(r=-0.89,P=0.02),和眼轴呈显著正相关(r=0.98,P=0.001)。各时间点自身对照组和正常对照组的各指标差异均无统计学意义(均为P>0.05)。结论 C57BL/6小鼠形觉剥夺性近视的发生和发展中存在后部巩膜组织F4/80+/CD206+巨噬细胞的上调,与屈光度和眼轴有相关性,提示F4/80+/CD206+巨噬细胞参与近视的发生发展过程。

Abstract:Objective To investigate the dynamic expression levels of F4/80+ and CD206+ macrophages in the posterior sclera in mice with form-deprivation myopia.Methods Twenty four C57BL/6 mice (48 eyes),4-week old,undergoing monocular form-deprivation (12 mice) and untreatment (12 mice),were divided into 3 groups according to random number table:form deprivation group (mice with monocular form-deprivation);self-control group (fellow eyes without form deprivation) and normal control group (no treatment).The refractive diopter of mice was examined before operation,on day 14 and 28.Then 6 mice were sacrificed,and the axial length,the expression levels of F4/80+ and CD206+ in the posterior sclera were examined by flow cytometry on day 14 and 28,respectively.The dynamic changes and internal relations of above parameters were investigated.Results There were no significant differences among the 3 groups in terms of refractive diopter (all P>0.05).On day 14,there were no significant differences in refractive diopter and axial length in the three groups (all P>0.05).In the form deprivation group,F4/80+ macrophages and the percentage of F4/80+,F4/80+/CD206+ macrophages were significantly higher than those of the self-control group (all P<0.05).At 28 d,form deprivation group showed significantly myopic shift (P=0.001) accompanied with elongated axial length (P=0.02) compared with self-control group;in the form deprivation group,F4/80+ macrophages and the percentage of F4/80+,F4/80+/CD206+ macrophages were significantly higher than those of the self-control group (all P<0.05).In the form deprivation group,F4/80+/CD206+ macrophages in mouse posterior sclera at 14 d was significantly higher compared with that of 28 d (P=0.02).The expression of F4/80+/CD206+ was negatively correlated with refractive diopter (r=-0.89,P=0.02) and positively correlated with axial length (r=0.98,P=0.001) on day 28.There were no significant differences in the indexes between the self-control group and the normal control group at each time point (all P>0.05).Conclusion The expression of F4/80+/CD206+ macrophage is shown to be up-regulated and correlated with refractive diopter and axial length in mouse posterior sclera in form-deprivation myopia,which implies that F4/80+/CD206+ macrophage maybe involves in the development of myopia.
