菌灵诱导小鼠睾丸支持细胞凋亡及周期阻滞的研究

摘    要:目的 观察多菌灵对小鼠睾丸支持细胞系(TM4)周期及凋亡的影响,并探讨其凋亡机制。方法 不同浓度(0.25、0.5、1、2、4、8 μg/ml)的多菌灵体外作用TM4细胞48 h,运用MTT法检测OD值及计算细胞存活率,PI染色法检测TM4细胞周期的分布变化,Annexin V/PI双标识标记法检测TM4细胞的凋亡情况,应用荧光探针DCFH - DA法检测TM4细胞内活性氧(Reactive Oxygen Species,ROS)的相对荧光强度,qRT - PCR法检测TM4细胞内Bax和Bcl - 2 mRNA的表达水平。结果 随着多菌灵浓度的升高,TM4细胞的增殖受到抑制,细胞周期分布发生变化,G2/M期的细胞呈现增加的趋势(P<0.01),胞内Bax mRNA和ROS表达水平也逐步升高(P<0.05,P<0.01),而Bcl - 2 mRNA表达则逐步下降(P<0.01)。结论 多菌灵可诱导TM4细胞凋亡并发生G2/M期阻滞,其作用机制可能与Bax、Bcl - 2 mRNA及ROS的异常表达有关。

Abstract:Objective The aim of this study was to investigate the effect of carbendazim on the cell cycle and apoptosis of mouse Sertoli cells (TM4) and its mechanism. Methods TM4 cells were separately treated with 0.25, 0.5, 1, 2, 4, 8 μg/ml of carbendazim solution for 48 h. Cell OD values and viability were measured by MTT method. After PI staining, the changes in cell cycle distribution were detected by flow cytometry. Apoptosis was analyzed using Annexin V/PI double binding assay. The relative fluorescence intensity of ROS in TM4 cells was measured by DCFH-DA method. And the expression of apoptosis related protein Bax and Bcl-2 mRNA was analyzed by real-time fluorescence quantitative PCR (qRT-PCR). Results Compared with the control group, with the increase of carbendazim concentration, the proliferation of TM4 cells was inhibited; the distribution of cell cycle changed and G2/M phase cells increased(P<0.01); the expression of intracellular Bax, mRNA and ROS were gradually increased (P<0.05,P<0.01), whereas the expression of Bcl-2 mRNA decreased(P<0.05). Conclusion Carbendazim was able to induce apoptosis of TM4 cells and cause G2/M phase arrest, and its mechanism might be related to the abnormal expression of Bax mRNA, Bcl-2 mRNA and ROS.
