| 摘 要： | 目的探讨血管内皮细胞生长因子（VEGF）对体外培养的脐静脉内皮细胞（HUVEC）增殖和迁移能力的影响。方法以VEGF基因转染HUVECs和在培养液中加入VEGF蛋白后培养HUVEC,用RT-PCR法检测VEGF mRNA的表达鉴定转染的效果,用3H-TdR掺入法检测VEGF对HUVECs增殖的影响,用划痕实验观察VEGF对HUVECs迁移能力的影响。结果转染VEGF基因的HUVECs中VEGFmRNA的表达高于对照组,表明转染成功。3H-TdR掺入法和划痕实验显示,转染VEGF基因和在培养液中加入VEGF蛋白,都可促进体外培养的HUVECs增殖和迁移。结论VEGF能够促进体外培养的HUVECs增殖和迁移;将VEGF基因导入细胞与直接在培养基中加入VEGF蛋白具有相同的效果。

| Abstract: | AIM To explore the effect of vascular endothelial growth factor (VEGF) on proliferation and migration ability of human umbilical vein endothelial cells (HUVECs) cultured in vitro. METHODS The HUVECs were transfected by VEGF gene and the VEGF protein was added into the culture fluid for HUVECs. The expression of VEGF mRNA was detected by RT-PCR to evaluate the efficacy of the transfection.
