| 摘 要： | 目的:观察独活寄生汤干预胶原性骨关节炎模型大鼠的作用机制。方法:选择50只SD大鼠,随机选取8只作为正常组,其余42只采用牛/鸡二型胶原诱导建立胶原性关节炎(CIA)模型,将复制成功的模型大鼠随机分为模型组、地塞米松组、独活寄生汤组。地塞米松组、独活寄生汤组大鼠每天分别给予地塞米松、独活寄生汤灌胃,正常组、模型组每天给予等体积蒸馏水灌胃。持续30 d后取材,通过测定不同实验组大鼠体重、关节指数、脾脏指数、血清白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平和踝关节病理组织检查,比较独活寄生汤对CIA模型大鼠的影响。结果:与正常组相比,模型组关节炎指数升高、足肿胀明显,差异有统计学意义(均P<0.05)。模型组大鼠血清IL-6、TNF-α水平高于正常组,差异有统计学意义(均P<0.01)。地塞米松组、独活寄生汤组大鼠关节炎指数、血清IL-6、TNF-α水平低于模型组,差异有统计学意义(均P<0.01)。HE染色后观察各组大鼠关节病理学变化,地塞米松组、独活寄生汤组大鼠细胞侵蚀及骨质侵蚀情况均轻于模型组,且未形成明显血管翳。结论:独活寄生汤通过抑制IL-6、TNF-α水平,减轻大鼠CIA症状。

| Abstract: | Objective:To observe effect of Duhuo Jisheng decoction on collagen osteoarthritis model rats.Methods:50 SD rats,8 were selected as the normal group,and the remaining 42 were induced by bovine/chicken type Ⅱ collagen to induce rat collagen arthritis(CIA)model.The successfully duplicated model rats were randomly divided into the model group,the dexamethasone group and the Duhuo Jisheng decoction group.The dexamethasone group were given dexamethasone,and the Duhuo Jisheng decoction group given Duhuo Jisheng decoction by gavage every day.The normal group and the model group were both given distilled water by gavage every day.The samples were collected after 30 days.Measuring body weight,joint index,spleen index,serum IL-6,TNF-α levels and ankle pathological examination in different experimental groups,to compare effect of Duhuo Jisheng decoction on CIA model rats.Results:Compared with the normal group,arthritis index and swelling foot of the model group were increased,difference statistically significant(all P<0.05).The serum levels of IL-6 and TNF-α in the model group were higher than those in the normal group,difference statistically significant(all P<0.01).The arthritis index,serum IL-6 and TNF-α of rats in the dexamethasone group and the Duhuo Jisheng decoction group were lower than those in the model group,difference statistically significant(all P<0.01).After HE staining,pathological changes of the joints of rats in each group were observed.The cell erosion and bone erosion of rats in the dexamethasone group and the Duhuo Jizhu decoction group were lighter than those in the model group,and no obvious pannus was formed.Conclusion:Duhuo Jisheng decoction can alleviate symptoms of CIA in rats by inhibiting the levels of inflammatory factors IL-6 and TNF-α. |
