Objective:To analyze the mechanism of antifibrotic role of Qigong Kangxian decoction (QKD) and the component herbs in rat models of hepatic fibrosis induced by immune injury. Method: A modified comparison method of serum pharmacology was applied to analyze, and QKD's impact on growth of hepatic stellate cell(HSC-T6) cell and expression of transforming growth factor (TGF-β1) receptor mRNA protein. Result: significant increase in cell growth of stellate cell(HSC-T6) were observed in the rat models comparing with that of normal healthy rats. While rat models treated with different QKD doses showed significant inhibitory effects on growth of HSC-T6 cell (P<0.01 or P<0.05) comparing with model rats. Comparing with normal control group, TGF-β1 mRNA concentration increased significantly in rat models (P<0.01), but in rat models treated with QKD, TGF-β1 mRNA concentration were all regulated to decrease obviously (P<0.01). Conclusion: One of the main mechanisms of antifibrotic role of QKD and individual herbs is the inhibitory impact on HSC cell growth, and down-regulation of TGF-β1mRNA receptor protein expression. QKD shows a dose dependent effect, which is better than that of Huoxue formula and Ruanjian formula.
